چکیده
Cytotoxic T lymphocyte (CTL) has been found to be activated by cutaneous dendritic cell (DC) in inducible skin-associated lymphoid tissue (iSALT) a murine contact hypersensitivity (CHS) model. Cutaneous DCs are classified into Langerhans (LC), conventional DC1 (cDC1), DC2 (cDC2) and monocyte-derived DC (MoDC); however, which subsets activate CTL within iSALT during the elicitation phase of CHS remained unclear. To clarify this, we used Langerin-diphtheria toxin receptor (DTR), XCR1-DTR CD301b-DTR mice deplete LC, cDC1 cDC2, respectively. remove MoDC, anti-CCR2 depleting antibody. Mice were sensitized applying 0.5% 2,4-dinitrofluorobenzene (DNFB) abdominal skin (day 0). Following administration diphtheria or antibody, elicited 0.3% DNFB ear 5). After 24 hours, swelling interferon-gamma (IFN-γ) production from significantly attenuated depletion cDC2 MoDC. However, there no significant differences LC cDC1. Immunohistochemical analysis revealed that cluster formation was suppressed but not In addition, colocalized with clusters. Taken together, these results suggest CHS. Further study elucidate molecular mechanism interaction between will provide new therapeutic strategies for CTL-mediated diseases.

متن کاملDendritic cell-glioma fusion activates T lymphocytes by elevating cytotoxic efficiency as an antitumor vaccine
BACKGROUND Hybrid cells produced by fusions of tumor and dendritic cells (DC) have demonstrated remarkable efficacy for priming the anti-tumor immune response. In the current study, we examined the antitumor activity of cytotoxic T lymphocytes (CTLs) primed in response to a tumor vaccine comprising a glioma-DC fusion as part of a therapeutic against glioma. MATERIAL AND METHODS Primary cultur...
متن کاملCell Surface Glycoproteins of Murine Cytotoxic T Lymphocytes
The immune system is endowed with great flexibility. It has at its disposal a large number of alternative pathways of reaction against foreign, immunogenic substances. A sizeable part of this impressive flexibility is due to the fact that a number of subsets of specialized cell types coexist and interact within this system. Among these cell types, lymphocytes have been shown to play a dominatin...
متن کاملcell surface glycoproteins of murine cytotoxic T lymphocytes. I. T 145, a new cell surface glycoprotein selectively expressed on Ly 1-2(+) cytotoxic T lymphocytes
T lymphocytes at various stages of maturation and differentiation have been isolated by cellular fractionation procedures and characterized by cell surface markers and functional assays, The cell surface glycoproteins of the various T-cell preparations have been selectively radiolabeled by the galactose oxidase-tritiated sodium borohydride technique and analyzed by sodium dodecyl sulfate-polyac...
